The SH 2 domain protein GRB 7 is co amplified , overexpressed and in a tight complex with HER 2 in breast cancer . ^^^ We find that GRB 7 is amplified in concert with HER 2 in several breast cancer cell lines and that GRB 7 is overexpressed in both cell lines and breast tumors . ^^^ GRB 7 , through its SH 2 domain , binds tightly to HER 2 such that a large fraction of the tyrosine phosphorylated HER 2 in SKBR 3 cells is bound to GRB 7 . ^^^ The fact that GRB 7 and HER 2 are both overexpressed and bound tightly together suggests that this basic signaling pathway is greatly amplified in certain breast cancers . . ^^^ |

Structural determinants of the interaction between the erbB 2 receptor and the Src homology 2 domain of Grb 7 . ^^^ The Src homology 2 ( SH 2 ) domain containing protein Grb 7 and the erbB 2 receptor tyrosine kinase are overexpressed in a subset of human breast cancers . ^^^ Whereas the Grb 7 SH2 domain binds strongly to erbB 2 , the SH 2 domain of Grb 14 , a protein closely related to Grb 7 , does not . ^^^ We have investigated the preferred binding site of Grb 7 within the erbB 2 intracellular domain and the SH 2 domain residues that determine the high affinity of Grb 7 compared with Grb 14 for this site . ^^^ Phosphopeptide competition and site directed mutagenesis revealed that Tyr 1139 of erbB 2 is the major binding site for the Grb 7 SH2 domain , indicating an overlap in binding specificity between the Grb 7 and Grb 2 SH2 domains . ^^^ |

Coamplification and coexpression of GRB 7 and ERBB 2 is found in high grade intraepithelial neoplasia and in invasive Barrett ' s carcinoma . ^^^ GRB 7 is localized in close proximity to the ERBB 2 gene within an amplicon previously identified in Barrett ' s adenocarcinoma ( BCA ) . ^^^ We evaluated gene amplification and mRNA expression of GRB 7 and ERBB 2 in Barrett ' s carcinoma and its associated precursor lesions to assess their possible role in Barrett ' s malignant transformation . ^^^ Copy number and expression levels were analyzed by Q PCR ( GRB 7 , ERBB 2 ) and QRT PCR ( GRB 7 , ERBB 2 ) using TaqMan technology and fluorescence in situ hybridization ( ERBB 2 ) in a series of 24 laser microdissected samples of Barrett ' s carcinomas and 32 samples of associated premalignant lesions . ^^^ Parallel analysis of gene copy number changes and expression levels demonstrated that GRB 7 and ERBB 2 displayed concomitant elevated expression levels and increased copy numbers in 32 % of Barrett ' s carcinomas . ^^^ |
