Association of csk homologous kinase ( CHK ) ( formerly MATK ) with HER 2 / ErbB 2 in breast cancer cells . ^^^ In this report , we investigated whether CHK is expressed in breast cancer tissues and whether it participates in the ErbB 2 signaling pathway in T47D and MCF 7 breast cancer cell lines . ^^^ To study the role of CHK in the ErbB 2 signaling pathway , glutathione S transferase fusion proteins containing the SH 2 and SH 3 domains of CHK were generated . ^^^ CHK SH 2 and CHK SH 3 SH2 , but not CHK SH 3 or CHK NH 2 SH3 , precipitated the tyrosine phosphorylated ErbB 2 upon stimulation with heregulin . ^^^ EGF or interleukin 6 stimulation of T47D cells failed to induce CHK SH 2 association with ErbB 2 , the EGF receptor , or the interleukin 6 receptor . ^^^

Recently , we observed that CHK is associated with the ErbB 2 / neu receptor upon heregulin stimulation of breast cancer cells . ^^^ Confocal microscopy analysis revealed co localization of CHK with ErbB 2 in these primary specimens ( 6 / 6 ) . ^^^ Phosphopeptide inhibition of the in vitro interaction of CHKSH 2 or native CHK with ErbB 2 / neu , as well as site directed mutagenesis of ErbB 2 / neu , indicated that CHKSH 2 binds to Tyr 1253 of ErbB 2 / neu . ^^^ Moreover , CHK was able to down regulate ErbB 2 / neu activated Src kinases . ^^^ These studies indicate that CHK binds , via its SH 2 domain , to Tyr 1253 of the activated ErbB 2 / neu and down regulates the ErbB 2 / neu mediated activation of Src kinases , thereby inhibiting breast cancer cell growth . ^^^

Csk homologous kinase ( CHK ) and ErbB 2 interactions are directly coupled with CHK negative growth regulatory function in breast cancer . ^^^ Our previous studies demonstrated that Csk homologous kinase ( CHK ) acts as a negative growth regulator of human breast cancer through inhibition of ErbB 2 / neu mediated Src family kinase activity ( Bougeret , C . , Jiang , S . , Keydar , I . , and Avraham , H . ( 2001 ) J . ^^^ The interaction between the CHK SH 2 domain and Tyr ( P ) ( 1248 ) of the ErbB 2 receptor has been shown to be specific and critical for CHK function . ^^^ In this report , we investigated whether the interaction of the CHK SH 2 domain and ErbB 2 is directly related to the inhibition of heregulin stimulated Src kinase activity . ^^^ These results indicate that the specific interaction of CHK and ErbB 2 via the SH 2 domain of CHK is directly related to the growth inhibitory effects of CHK . ^^^
