We analyzed apolipoprotein E ( apo E ) genotypes in 53 Japanese sporadic Creutzfeldt Jakob disease ( CJD ) patients and 100 normal controls using the polymerase chain reaction ( PCR ) and restriction fragment length polymorphism ( RFLP ) methods . ^^^ The apo E allelic frequencies in Japanese sporadic CJD patients and our control population were as follows : epsilon 2 , 6 . 6 % versus 7 . 5 % ; epsilon 3 , 82 . 1 % versus 81 . 5 % ; epsilon 4 , 11 . 3 % versus 11 . 0 % . ^^^ These results indicate that there is no association between apo E genotype and sporadic CJD in Japan . . ^^^

We determined codon 129 of the prion gene and the Apo E variants in Creutzfeldt Jakob disease ( CJD ) using PCR and restriction digest . ^^^ We found a significant correlation between valine at codon 129 of the prion protein gene and the presence of plaque in CJD and a later age of onset in CJD cases possessing the Apo E 2 allele . ^^^

OBJECTIVE : To analyze the distribution of apolipoprotein E ( Apo E ) phenotypes between patients with Creutzfeldt Jakob disease ( CJD ) and control subjects . ^^^ Patients with definite CJD had a mean ( SD ) Apo E concentration of 3 . 4 ( 2 . 0 ) mg / L ; patients with probable CJD , 3 . 1 ( 1 . 6 ) mg / L ; patients with possible CJD , 3 . 8 ( 2 . 2 ) mg / L ; and subjects with other diseases , 3 . 0 ( 1 . 7 ) mg / L . ^^^ CONCLUSIONS : We found no significant difference in the distribution of Apo E phenotypes between patients with CJD and controls . ^^^ The Apo E phenotype had no influence on the duration of CJD . ^^^ Our data do not support an association of Apo E 4 with either the duration or time at onset of CJD . . ^^^

In the present study we have compared the interaction of apoE with A beta , the gelsolin derived amyloid fragment AGel ( 183 210 ) and the amyloidogenic prion fragments PrP ( 109 122 ) and PrP ( 109 141 ) . ^^^ We show that , similar to A beta , also AGel and PrP fragments can form a complex with apoE , and that the interaction between apoE and the amyloidogenic protein fragments is mediated through the same binding site on apoE . ^^^ We also show that apoE increases the thioflavin T fluorescence of PrP and AGel and that apoE influences the content of beta sheet conformation of these amyloidogenic fragments . ^^^
