Differential effects of the hsp 70 binding protein BAG 1 on glucocorticoid receptor folding by the hsp 90 based chaperone machinery . ^^^ Inasmuch as BAG 1 has been found in association with several members of the steroid receptor family , we have examined the effect of BAG 1 on GR folding and GR . hsp90 heterocomplex assembly . ^^^ BAG 1 was present in reticulocyte lysate at a BAG 1 : hsp70 / hsc70 molar ratio of approximately 0 . 03 , and its elimination by immunoadsorption did not affect GR folding and GR . hsp 90 heterocomplex assembly . ^^^ At low BAG 1 : hsp70 / hsc70 ratios , BAG 1 promoted the release of Hop from the hsp 90 based chaperone system without inhibiting GR . hsp90 heterocomplex assembly . ^^^ However , at molar ratios approaching stoichiometry with hsp 70 , BAG 1 produced a concentration dependent inhibition of GR folding to the steroid binding form with corresponding inhibition of GR . hsp90 heterocomplex assembly by the minimal five protein chaperone system . ^^^

Available data on the negative regulation of glucocorticoid receptor ( GR ) action by the BAG 1 proteins identify two modes of action : inhibition of the hormone binding activity of the GR and a more direct nuclear action at the level of regulation of the transactivation function of the receptor . ^^^ In the latter case , the BAG 1 proteins repress DNA binding by the GR in a process that requires prior binding of Hsp70 / Hsc70 to the receptor . ^^^

The anti apoptotic , glucocorticoid receptor cochaperone protein BAG 1 is a long term target for the actions of mood stabilizers . ^^^ Because BAG 1 attenuates glucocorticoid receptor ( GR ) nuclear translocation , activates ERK ( extracellular signal regulated kinase ) MAP ( mitogen activated protein ) kinases , and potentiates anti apoptotic functions of BCL 2 , extensive additional studies were undertaken . ^^^ Furthermore , small interfering RNA studies showed that these inhibitory effects on GR activity were mediated , at least in part , through BAG 1 . ^^^ Additionally , these studies suggest that regulation of GR mediated plasticity may play a role in the treatment of bipolar disorder and raise the possibility that agents affecting BAG 1 more directly may represent novel therapies for this devastating illness . . ^^^
