One protein that co occupies the distal nGRE in vitro along with GR is the POU domain transcription factor Oct 1 . ^^^ Thus , the tethering of GR to the GnRH distal nGRE , by virtue of a direct or indirect association with DNA bound Oct 1 , could play a role in hormone dependent transcriptional repression of the GnRH gene . ^^^ In contrast , Oct 1 does not appear to be a component of the GR containing protein complex that is bound to the proximal nGRE . . ^^^

GR also exerts negative actions on transcription , e . g . , by interfering with the function of several other transcription factors such as AP 1 , NK kappa B , CREB , and Oct 1 . ^^^

Remarkably , we found little or no relationship between the spatial distribution of the glucocorticoid receptor , Oct 1 and E2F 1 on the one hand and RNA polymerase 2 and transcription sites on the other hand . ^^^

Addition of in vitro translated glucocorticoid receptor DNA binding domain to the nuclear extract prevented Oct 1 and Pbx from binding to the PRL element . ^^^

The glucocorticoid receptor is tethered to DNA bound Oct 1 at the mouse gonadotropin releasing hormone distal negative glucocorticoid response element . ^^^ In addition , the interaction of purified GR with DNA bound Oct 1 is altered when Oct 1 is bound to the consensus Oct 1 site . ^^^

Transcriptional synergism between glucocorticoid receptor ( GR ) and octamer transcription factors 1 and 2 ( Oct 1 and Oct 2 ) in the induction of mouse mammary tumor virus ( MMTV ) transcription has been proposed to be mediated through directed recruitment of the octamer factors to their binding sites in the viral long terminal repeat . ^^^ However , physiologically relevant binding to the POU domain appeared to be a property restricted to only a few nuclear receptors as only GR , progesterone receptor ( PR ) , and androgen receptor ( AR ) , were found to interact physically and functionally with Oct 1 and Oct 2 in transfected cells . ^^^

DNA dependent conformational changes in Oct 1 play a major role in recruiting GR to the distal nGRE and impacts transcriptional repression brought about by either glucocorticoids or tumor promoting phorbol esters . ^^^

Gel shift assays using nuclear extracts of U 251MG cells demonstrated that RE 1a contains binding sites for transcription factors Sp 1 , the glucocorticoid receptor , and the E box binding factors , whereas RE 1b contains a binding site for the octamer motif binding protein ( OCT 1 / OCT 2 ) . ^^^

All of these factors ( GR , c Jun , OCT 1 ) have transcription activation domains , but STAT 3 is required for the observed transcriptional increase . ^^^

Here we demonstrate that specific and concomitant binding of nuclear factor 1 ( NF 1 ) and octamer factor 1 ( Oct 1 ) to their cognate sites within the MMTV promoter induce a partial nucleosome positioning that is an intermediary state between the randomly organized inactive promoter and the hormone and GR activated promoter containing distinctly positioned nucleosomes . ^^^ The NF 1 and Oct 1 binding also facilitate hormone dependent GR DNA interaction and result in a faster and stronger hormone response . ^^^

Functional interference between the ubiquitous and constitutive octamer transcription factor 1 ( OTF 1 ) and the glucocorticoid receptor by direct protein protein interaction involving the homeo subdomain of OTF 1 . ^^^ Here we present evidence that the glucocorticoid receptor can negatively regulate OTF 1 function by a mechanism that is independent of DNA binding . ^^^ In vitro , the DNA binding activity of partially purified OTF 1 was repressed following incubation with purified glucocorticoid receptor . ^^^ Finally , preliminary evidence indicates that the homeo subdomain of OTF 1 that directs formation of a complex with VP 16 may also be critical for interaction with the glucocorticoid receptor . ^^^ Moreover , the functional interference between OTF 1 and the glucocorticoid receptor represents a novel regulatory mechanism in the cross coupling of signal transduction pathways of nuclear receptors and constitutive transcription factors . . ^^^

In this study , we investigated the hormone dependent interaction between the DNA binding domain ( DBD ) of the glucocorticoid receptor ( GR ) and the POU domains of octamer transcription factors 1 and 2 ( Oct 1 and Oct 2 , respectively ) . ^^^ Our results indicate that the GR DBD binds directly , not only to the homeodomains of Oct 1 and Oct 2 but also to the homeodomains of several other homeodomain proteins . ^^^
