A novel ligand of the formyl peptide receptor : annexin 1 regulates neutrophil extravasation by interacting with the FPR . ^^^ Here we show that annexin 1 acts through the formyl peptide receptor ( FPR ) on human neutrophils . ^^^ Peptides derived from the unique N terminal domain of annexin 1 serve as FPR ligands and trigger different signaling pathways in a dose dependent manner . ^^^ These findings identify annexin 1 peptides as novel , endogenous FPR ligands and establish a mechanistic basis of annexin 1 mediated antiinflammatory effects . . ^^^

In a model of mouse peritonitis , FPR antagonists abrogated the anti migratory effects of peptides Ac 2 26 and Ac 2 12 , with a partial reduction in annexin 1 effects . ^^^ Binding of annexin 1 to circulating leukocytes was reduced ( > 50 % ) in FPR KO mice . ^^^ Finally , evidence of direct annexin 1 binding to murine FPR was obtained with HEK 293 cells transfected with the receptor . ^^^ Overall , these results indicate a functional role for FPR in the anti migratory effect of annexin 1 and derived peptides . . ^^^

We examined the 1 ) amino acid region of ANXA 1 that retained the protective effect in a model of rat heart IR ; 2 ) changes in endogenous ANXA 1 in relation to the IR induced damage and after pharmacological modulation ; and 3 ) potential involvement of the formyl peptide receptor ( FPR ) in the protective action displayed by ANXA 1 peptides . ^^^ Finally , an antagonist to the FPR receptor selectively inhibited the protective action of peptide ANXA 1 and its derived peptides against IR injury . ^^^

In this study we show that a peptide derived from the N terminal domain of the anti inflammatory protein annexin 1 ( lipocortin 1 ) can activate all three FPR family members at similar concentrations . ^^^ The annexin 1 peptide initiates chemotactic responses in human monocytes that express all three FPR family members and also desensitizes the cells toward subsequent stimulation with bacterial peptide agonists . ^^^ Experiments using HEK 293 cells stably expressing a single FPR family member reveal that all three receptors can be activated and desensitized by the N terminal annexin 1 peptide . ^^^ These observations identify the annexin 1 peptide as the first endogenous ligand of FPRL 2 and indicate that annexin 1 participates in regulating leukocyte emigration into inflamed tissue by activating and desensitizing different receptors of the FPR family . . ^^^

Recent interest in the annexin 1 field has come from the notion that specific G protein coupled receptors , members of the formyl peptide receptor ( FPR ) family , appear to mediate the anti inflammatory actions of this endogenous mediator . ^^^ Similarly , peptide Ac 2 26 inhibition of specific markers of heart injury ( specifically myeloperoxidase activity , CXC chemokine KC contents , and endogenous annexin 1 protein expression ) was virtually identical in heart samples collected from wild type and FPR null mice . ^^^ Thus , this study sheds light on the receptor mechanism ( s ) mediating annexin 1 induced cardioprotection and shows a pivotal role for ALX and circulating neutrophil , whereas it excludes any functional involvement of mouse FPR . ^^^
