Human platelet derived transforming growth factor beta ( TGF beta 1 ) increases the accumulation of the extracellular matrix proteins , fibronectin and type 1 collagen , in mesenchymal and epithelial cells . ^^^ To determine the basis for this effect , we have examined the levels of mRNAs corresponding to fibronectin and alpha 2 ( 1 ) procollagen in NRK 49 rat fibroblasts and L6E9 rat myoblasts treated with TGF beta 1 . ^^^ The increase in fibronectin and alpha 2 ( 1 ) procollagen mRNAs was detectable 2 h after addition of TGF beta 1 to the cells and their maximal levels remained constant for several days . ^^^ Actinomycin D , but not cycloheximide , inhibited the increase in fibronectin and alpha 2 ( 1 ) procollagen mRNA levels induced by TGF beta 1 . ^^^ The results indicate that TGF beta 1 controls the composition and abundance of extracellular matrices at least in part by inducing a coordinate increase in the levels of fibronectin and type 1 collagen mRNAs . . ^^^ |

Antibodies against TGF beta 1 or FN almost completely abolished the chemotactic migration of B 16 BL6 cells to the CM ( B 16 BL6 / fibroblast ) or CM ( TGF beta 1 treated fibroblast ) when these antibodies were co incubated with fibroblasts and either B 16 BL6 or TGF beta 1 . ^^^ Analysis of amounts of TGF beta 1 or FN in various CM using ELISA plates , and using their specific antibodies , revealed that the concentration of TGF beta 1 in the CM ( B 16 BL6 ) was slightly higher than in the CM ( B 16 F1 ) , and the amount of FN in the CM ( B 16 BL6 / fibroblast ) was twice as high as in the CM ( B 16 F1 / fibroblast ) . ^^^ These results suggest that TGF beta 1 released from B 16 BL6 cells can stimulate fibroblasts to produce FN ; consequently , the tumor cells were able to chemotactically migrate toward the released FN , and the differences in invasive and migratory activities towards fibroblasts in B 16 BL6 and B 16 F1 cells may in part be due to the amounts of TGF beta 1 from tumor cells and of FN from TGF beta 1 stimulated fibroblasts . . ^^^ |

To investigate the relationship between plasminogen activator ( PA ) , plasminogen activator inhibitor 1 ( PAI 1 ) and the extracellular matrix in malignant and normal lung epithelial cells and to determine whether malignant lung epithelial cells may be more invasive than normal lung epithelial cells because of differences in expression of these proteins in response to TGF beta , the regulation of PA , PAI 1 , fibronectin , laminin and thrombospondin by TGF beta 1 in human non small cell lung cancer ( NSCLC ) cells was examined and compared with normal human bronchial epithelial ( NHBE ) cells . ^^^ Treatment of NSCLC cells with TGF beta 1 resulted in a persistent increase in expression of the mRNAs for fibronectin , laminin and thrombospondin ; expression of fibronectin protein also increased after treatment with TGF beta 1 in these cells . ^^^ When NHBE cells were similarly cultured in the presence of TGF beta 1 , expression of fibronectin mRNA also increased in a persistent manner ; however , only an early transient increase in the level of the mRNAs for laminin and thrombospondin was detected in these cells . ^^^ These data show that there is differential regulation of the genes for PA and PAI 1 and the extracellular matrix protein fibronectin in response to TGF beta 1 not only when NSCLC and NHBE cells are compared , but also when different NSCLC cells are compared with each other . . ^^^ |
