Interactions of the DNA mismatch repair proteins MLH 1 and MSH 2 with c MYC and MAX . ^^^ As well as known partners of MLH 1 such as PMS 1 , MLH 3 and MBD 4 , we identified the carboxy terminus of the human c MYC proto oncogene as an interacting sequence . ^^^ We demonstrate , both in vitro by yeast two hybrid and GST fusion pull down experiments , as well as in vivo by coimmunoprecipitation from human tumour cell extracts , that MLH 1 interacts with the c MYC protein . ^^^

Myc down regulation sensitizes melanoma cells to radiotherapy by inhibiting MLH 1 and MSH 2 mismatch repair proteins . ^^^ CONCLUSIONS : Data herein reported show that Myc down regulation lowers the apoptotic threshold in melanoma cells by inhibiting MLH 1 and MSH 2 proteins , thus increasing cell sensitivity to gamma radiation in a p 53 independent fashion . ^^^
