Bifunctional transcriptional properties of YY 1 in regulating muscle actin and c myc gene expression during myogenesis . ^^^ The c myc and skeletal alpha actin gene promoters contain YY 1 binding sites thought to act either as positive or negative cis acting elements . ^^^ Through manipulating the intracellular level of YY 1 by treating primary myoblasts with bromodeoxyuridine ( BrdU ) , which inhibited myogenesis and increased the YY 1 protein content , or by transfecting YY 1 cDNA expression vector , we show that it can simultaneously inhibit and activate expression of the skeletal alpha actin and c myc genes , respectively . ^^^ We propose that the high level of YY 1 in proliferating myoblasts might serve to maintain c myc expression and suppress muscle actin expression , which can then be gradually reversed by downregulating YY 1 activity toward myogenesis . . ^^^ |

YY 1 can inhibit c Myc function through a mechanism requiring DNA binding of YY 1 but neither its transactivation domain nor direct interaction with c Myc . ^^^ The proto oncoprotein c Myc and the multifunctional transcriptional regulator YY 1 have been shown previously to interact directly in a manner that excludes Max from the complex ( Shrivastava et al . , 1993 ) . ^^^ As binding to Max is necessary for all known c Myc activities we have analysed the influence of YY 1 on c Myc function . ^^^ We demonstrate that YY 1 is a potent inhibitor of c Myc transforming activity . ^^^ The region in YY 1 required for inhibition corresponds to a functional DNA binding domain and is distinct from the domains necessary for direct binding to c Myc . ^^^ |

The c Myc oncoprotein is important in control of cellular proliferation and differentiation , while Yin Yang 1 ( YY 1 ) has been shown to control the expression of a number of cellular and viral genes . ^^^ In this study , we show that USF / c Myc up regulates , while YY 1 down regulates the promoter activity of CXCR 4 , a coreceptor for T cell tropic HIV 1 entry . ^^^ Mutation of the E box abolished USF / c Myc mediated up regulation of CXCR 4 promoter activity , and mutation of the YY 1 binding site was associated with unresponsiveness to YY 1 mediated inhibition . ^^^ These data suggest that USF / c Myc and YY 1 may play an important role in the HIV 1 replicative cycle , by modulating both the viral fusion / entry process and viral expression . . ^^^ |
