| Elevation of intracellular casein kinase 2 ( CKII ) levels through microinjection of purified CKII results in the rapid and transient induction of c fos in quiescent rat embryo fibroblasts , and activation of quiescent cells by serum is accompanied by the nuclear relocation of endogenous CKII . ^^^ The induction of c fos by CKII is inhibited by coinjection of oligonucleotides corresponding to the sequence of the serum response element ( SRE ) present in the c fos promoter , indicating that competitive displacement of positive factors from the endogenous c fos SRE prevents c fos induction by CKII . ^^^ Furthermore , the expression of c fos induced by either CKII injection or serum activation is also inhibited by microinjection of antibodies against the 67 kDa serum response factor ( p67SRF ) indicating the absolute requirement of p67SRF in this process . ^^^ Together , these data strongly support that CKII induces c fos expression through binding / activation of the phosphorylated p67SRF at the SRE sequence . . ^^^ |

| Since c fos transcription is induced by a number of the same agents that stimulate CKII activity , and since fos activation and CKII stimulation demonstrate similar rapid kinetics , a role was proposed for CKII in regulating fos expression via its phosphorylation of SRF . ^^^ However , when overexpressed stably in cells this mutant had no effect on endogenous c fos expression , suggesting that CKII phosphorylation of SRF is not the limiting event for fos activation . . ^^^ |

| We found that transactivation of the mdm 2 , p 21 ( WAF1 / CIP1 ) and cyclin G promoter was inhibited in three different cell lines whereas transactivation of the bax promoter was not affected in COS 1 cells but down regulated in MCO 1 and SaosS138V21 cells . p 53 mediated transrepression of the fos promoter was not influenced by coexpression of the CK 2 beta subunit . ^^^ |
