Mitofusins Mfn 1 and Mfn 2 coordinately regulate mitochondrial fusion and are essential for embryonic development . ^^^ The mitofusins , Mfn 1 and Mfn 2 , have been shown to affect mitochondrial morphology when overexpressed . ^^^ We find that mice deficient in either Mfn 1 or Mfn 2 die in midgestation . ^^^ However , whereas Mfn 2 mutant embryos have a specific and severe disruption of the placental trophoblast giant cell layer , Mfn 1 deficient giant cells are normal . ^^^ Embryonic fibroblasts lacking Mfn 1 or Mfn 2 display distinct types of fragmented mitochondria , a phenotype we determine to be due to a severe reduction in mitochondrial fusion . ^^^ |

The nuclear encoded mitochondrial proteins Mfn 1 and Mfn 2 are human homologues of the only known protein mediators of mitochondrial fusion , the Drosophila Fzo GTPase and Saccharomyces cerevisiae yFzo1p . ^^^ Although the Mfn 1 and Mfn 2 genes were broadly expressed , the two genes showed different levels of mRNA expression in different tissues . ^^^ Two Mfn 1 transcripts were detected at similar levels in a variety of human tissues and were dramatically elevated in heart , while Mfn 2 mRNA was abundantly expressed in heart and muscle tissue but present only at low levels in many other tissues . ^^^ |
