We found that AKAP 4 binds AKAP 3 and two novel spermatogenic cell specific proteins , Fibrous Sheath Interacting Proteins 1 and 2 ( FSIP 1 , FSIP 2 ) . ^^^ Transcription of Akap 4 , Akap 3 , and Fsip 1 begins in early spermatid development , whereas transcription of Fsip 2 begins in late spermatocyte development . ^^^ The AKAP 4 precursor is processed in the flagellum and only the mature form of AKAP 4 appears to bind AKAP 3 . ^^^ These results suggest that AKAP 3 is involved in organizing the basic structure of the fibrous sheath , whereas AKAP 4 has a major role in completing fibrous sheath assembly . . ^^^

Molecular genetic analysis of two human sperm fibrous sheath proteins , AKAP 4 and AKAP 3 , in men with dysplasia of the fibrous sheath . ^^^ Human AKAP 4 and AKAP 3 are structural proteins of the fibrous sheath that also function to anchor protein kinase A to this structure via the regulatory subunit of the kinase . ^^^ We hypothesized that defects in either AKAP 4 or AKAP 3 might cause DFS . ^^^ No quantitative or qualitative differences between patients with DFS and normal controls were detected when sperm proteins were analyzed by either silver staining or immunoblot analysis using antibodies raised against AKAP 4 and AKAP 3 . ^^^ Additionally , AKAP 4 and AKAP 3 from DFS sperm retained the ability to bind the regulatory subunit of protein kinase A . ^^^

RESULTS : Sp 17 is present in the head and tail of spermatozoa , in the tail it is in the fibrous sheath , which contains AKAP 3 and AKAP 4 . ^^^ Recombinant AKAP 3 and AKAP 4 RII binding domains were synthesized as glutathione S transferase ( GST ) fusion proteins immobilized on glutathione agarose resin and added to CHAPS extracts of human spermatozoa . ^^^ Western blots of bound and eluted proteins probed with anti Sp 17 revealed that AKAP 3 bound and precipitated a significant level of Sp 17 while AKAP 4 did not . ^^^ AKAP 4 binds AKAP 3 and expression of AKAP 3 is reduced in AKAP 4 knockout sperm , therefore we tested AKAP 4 knockout spermatozoa for Sp 17 and found that there was a reduction in the amount of Sp 17 expressed when compared to wild type spermatozoa . ^^^

We report the case of an infertile man with normal sperm count and total sperm immotility in which dysplasia of the fibrous sheath , Akap 3 , Akap 4 gene deletions , meiotic segregation of chromosomes 18 , 10 and Y and Y microdeletions were investigated . ^^^ PCR analysis revealed intragenic deletions of the Akap 3 and Akap 4 genes . ^^^ CONCLUSION : In this infertile patient , our results suggest a possible relationship between dysplasia of the fibrous sheath , partial deletions in the Akap 3 and Akap 4 genes and absence of AKAP 4 protein in the fibrous sheath . ^^^
