Utilizing a modified yeast two hybrid approach , we have identified the tumor susceptibility gene 101 ( TSG 101 ) and the vitamin D receptor interacting protein 150 ( DRIP 150 ) as proteins that interact specifically with a functional GR AF 1 surface . ^^^ In yeast and mammalian cells , TSG 101 represses whereas DRIP 150 enhances GR AF 1 mediated transactivation . ^^^ In addition , we show that another member of the DRIP complex , DRIP 205 , interacts with the GR ligand binding domain in a hormone dependent manner and facilitates GR transactivation in concert with DRIP 150 . ^^^ These results suggest that DRIP 150 and DRIP 205 functionally link GR AF 1 and AF 2 , and represent important mediators of GR transcriptional enhancement . . ^^^ |

MED 14 and MED 1 differentially regulate target specific gene activation by the glucocorticoid receptor . ^^^ The Mediator subunits MED 14 and MED 1 have been implicated in transcriptional regulation by the glucocorticoid receptor ( GR ) by acting through its activation functions 1 and 2 . ^^^ To understand the contribution of these Mediator subunits to GR gene specific regulation , we reduced the levels of MED 14 and MED 1 using small interfering RNAs in U2OS hGR osteosarcoma cells and examined the mRNA induction by dexamethasone of four primary GR target genes , interferon regulatory factor 8 ( IRF 8 ) , ladinin 1 , IGF binding protein 1 ( IGFBP 1 ) , and glucocorticoid inducible leucine zipper ( GILZ ) . ^^^ We found that the GR target genes differed in their requirements for MED 1 and MED 14 . ^^^ GR dependent mRNA expression of ladinin 1 and IRF 8 required both MED 1 and MED 14 , whereas induction of IGFBP 1 mRNA by the receptor was dependent upon MED 14 , but not MED 1 . ^^^ |
