Association of DAP 12 with activating CD94 / NKG2C NK cell receptors . ^^^ CD94 / NKG2C , an activating NK cell receptor of the C type lectin superfamily that binds to HLA E , noncovalently associates with DAP 12 , a membrane receptor containing an immunoreceptor tyrosine based activating motif . ^^^ Efficient expression of CD94 / NKG2C on the cell surface requires the presence of DAP 12 , and charged residues in the transmembrane domains of DAP 12 and NKG2C are necessary for this interaction . ^^^

NKG2A / B contains an immunoreceptor tyrosine based inhibitory motif ( ITIM ) and accordingly functions as an inhibitory receptor , whereas NKG2C and E / H associate with DAP 12 via a positively charged residue in their transmembrane domains and function as activation receptors . ^^^ NKG2F could be a receptor component with an as yet unidentified partner ( s ) , could function to regulate cell activation through competition for DAP 12 with other receptors , such as NKG2C and E / H , or it could simply be a vestigial gene product . . ^^^

We demonstrate that , in the absence of DAP 12 , rhesus monkey NKG2A is preferentially expressed at the cell surface with CD 94 due to a single amino acid difference in the transmembrane of NKG2A and NKG2C . ^^^ In the presence of DAP 12 , the ability of NKG2C to compete for cell surface CD 94 heterodimerization is enhanced and approaches that of NKG2A . ^^^
