RT PCR studies of MAP3K gene expression in RA and osteoarthritis synovial tissue demonstrated mitogen activated protein kinase / ERK kinase kinase ( MEKK ) 1 , MEKK 2 , apoptosis signal regulating kinase 1 , TGF beta activated kinase 1 ( TAK 1 ) gene expression while only trace amounts of MEKK 3 , MEKK 4 , and MLK 3 mRNA were detected . ^^^ Western blot analysis demonstrated immunoreactive MEKK 2 , TAK 1 , and trace amounts of MEKK 3 but not MEKK 1 or apoptosis signal regulating kinase 1 . ^^^

TAK 1 is recruited to the tumor necrosis factor alpha ( TNF alpha ) receptor 1 complex in a receptor interacting protein ( RIP ) dependent manner and cooperates with MEKK 3 leading to NF kappaB activation . ^^^ Interestingly , we also find that TAK 1 and MEKK 3 form a functional complex , in which TAK 1 regulates autophosphorylation of MEKK 3 . ^^^ The TAK 1 mediated regulation of MEKK 3 phosphorylation is dependent on the kinase activity of TAK 1 . ^^^ Although TAK 1 MEKK3 interaction is not affected by overexpressed TAB 1 , TAB 1 is required for TAK 1 activation and subsequent MEKK 3 phosphorylation . ^^^ Together , we conclude that TAK 1 is recruited to the TNF R 1 complex via RIP and likely cooperates with MEKK 3 to activate NF kappaB in TNF alpha signaling . . ^^^
