Here we report the identification and characterization of a novel protein designated TAB 3 , a TAB 2 like molecule that associates with TAK 1 and can activate NF kappaB similar to TAB 2 . ^^^ Cotransfection of siRNAs directed against both TAB 2 and TAB 3 inhibit both IL 1 and TNF induced activation of TAK 1 and NF kappaB . ^^^ These results suggest that TAB 2 and TAB 3 function redundantly as mediators of TAK 1 activation in IL 1 and TNF signal transduction . . ^^^ |

The activation of NF kappaB and IKK requires an upstream kinase complex consisting of TAK 1 and adaptor proteins such as TAB 1 , TAB 2 , or TAB 3 . ^^^ Mutations of the ZnF domain abolish the ability of TAB 2 and TAB 3 to bind polyubiquitin chains , as well as their ability to activate TAK 1 and IKK . ^^^ Significantly , replacement of the ZnF domain with a heterologous ubiquitin binding domain restored the ability of TAB 2 and TAB 3 to activate TAK 1 and IKK . ^^^ |
