IL 1beta and its downstream mediator TAK 1 inhibit SMAD 3 mediated TGFbeta target gene activation , whereas SMAD 3 nuclear translocation and DNA binding in response to TGFbeta are not affected . ^^^ IL 1beta transiently induces association between TAK 1 and the MAD homology 2 domain of SMAD 3 , resulting in SMAD 3 phosphorylation . ^^^ In conclusion , our data provide evidence for the existence of a direct inhibitory effect of the IL 1beta TAK 1 pathway on SMAD 3 mediated TGFbeta signaling , resulting in reduced TGFbeta target gene activation and restored proliferation of hematopoietic progenitors . . ^^^

This study used 4 . 7 kb of the ovine FSHbeta promoter linked to luciferase ( oFSHbetaLuc ) plus a well characterized activin responsive construct , p3TPLuc , to investigate the hypothesis that Smad 3 , TGFbeta activated kinase 1 ( TAK 1 ) , or both cause activin mediated induction of FSH . ^^^ Overexpression of either Smad 3 or TAK 1 induced oFSHbetaLuc in gonadotrope derived LbetaT 2 cells as much as activin itself . ^^^ In conclusion , the data presented here indicate that activation of TAK 1 ( and probably p 38 MAPK ) , but not Smad 3 , is necessary for triggering induction of oFSHbeta by activin . . ^^^
