Poly ( 1 C ) induced Toll like receptor 3 ( TLR 3 ) mediated activation of NFkappa B and MAP kinase is through an interleukin 1 receptor associated kinase ( IRAK ) independent pathway employing the signaling components TLR 3 TRAF6 TAK 1 TAB2 PKR . ^^^ Upon binding of dsRNA to TLR 3 , TRAF 6 , TAK 1 , and TAB 2 are recruited to the receptor to form a complex , which then translocates to the cytosol where TAK 1 is phosphorylated and activated . ^^^ Kinase inactive mutants of TAK 1 ( TAK1DN ) and PKR ( PKRDN ) inhibit poly ( dI . dC ) induced TLR 3 mediated NFkappaB activation , suggesting that both of these kinases play important roles in this pathway . . ^^^ |

Using embryonic fibroblasts lacking TAK 1 , TAB 1 , or TAB 2 , we have found that TNFR 1 , IL 1R , TLR 3 , and TLR 4 mediated NF kappaB and AP 1 activation are severely impaired in Tak 1 ( m / m ) cells , but they are normal in Tab 1 ( / ) and Tab 2 ( / ) cells . ^^^ |
