Using immunostaining techniques , we show that MCT 4 localizes with its accessory protein CD 147 in the basolateral membrane of rat thyroid follicular cells . 0.77986865^^^

MCT 3 , which is expressed in the basolateral membrane of the RPE wild type mouse , was expressed at very low levels in both the apical and basolateral membranes of the Bsg ( / ) mouse . ^^^ Western blot analysis of detergent soluble lysates prepared from wild type and Bsg ( / ) eyes confirmed that the levels of MCT 1 , MCT 3 , and MCT 4 protein were severely reduced in Bsg ( / ) mice . ^^^

Proton coupled monocarboxylate transporters ( MCT ) MCT 1 , MCT 3 , and MCT 4 form heterodimeric complexes with the cell surface glycoprotein CD 147 and exhibit tissue specific polarized distributions that are essential for maintaining lactate and pH homeostasis . ^^^ In contrast , MCT 3 and MCT 4 harbor dominant sorting information that cotargets CD 147 to the basolateral membrane in both epithelia . ^^^

However , in other cells MCT 1 and MCT 4 are intimately associated with an ancillary protein OX 47 ( also known as CD 147 ) . ^^^

In addition , MCT 1 and MCT 4 have been shown to interact specifically with OX 47 ( CD 147 ) , a member of the immunoglobulin superfamily with a single transmembrane helix . ^^^

CD 147 is tightly associated with lactate transporters MCT 1 and MCT 4 and facilitates their cell surface expression . ^^^ Here we use co immunoprecipitation and chemical cross linking to demonstrate that CD 147 specifically interacts with MCT 1 and MCT 4 , two members of the proton linked monocarboxylate ( lactate ) transporter family that play a fundamental role in metabolism , but not with MCT 2 . ^^^ In mammalian cell lines , expression is uniform but cross linking with anti CD 147 antibodies caused MCT 1 , MCT 4 and CD 147 , but not GLUT 1 or MCT 2 , to redistribute together into ' caps ' . ^^^ In MCT transfected cells , expressed protein accumulated in a perinuclear compartment , whereas co transfection with CD 147 enabled expression of active MCT 1 or MCT 4 , but not MCT 2 , in the plasma membrane . ^^^ We conclude that CD 147 facilitates proper expression of MCT 1 and MCT 4 at the cell surface , where they remain tightly bound to each other . ^^^

The monocarboxylate ( lactate ) transporters MCT 1 and MCT 4 require the membrane spanning glycoprotein CD 147 for their correct plasma membrane expression and function . ^^^

Human and mouse genetic expression of MCT members 1 , 2 , 3 , 4 and basigin , a chaperone protein of MCT 1 and MCT 4 , was qualitatively analysed using the reverse transcription nested polymerase chain reaction ( RT nested PCR ) in immature oocytes ( germinal vesicle stage ; GV ) , in non fertilised metaphase 2 ( MII ) oocytes and in embryos from 2 cell stage to blastocysts . ^^^

MCT 1 and MCT 4 require a monotopic ancillary protein , CD 147 , for expression of functional protein at the plasma membrane . ^^^

Post translational regulation of MCT 1 and MCT 4 occurs , in part , by interaction with CD 147 , an accessory protein that is necessary for trafficking , localization , and functional expression of these transporters . ^^^

CD 147 , a chaperone protein for MCT 1 and MCT 4 , was equally expressed by both membranes . ^^^

Translocation of monocarboxylate transporters MCT 1 and MCT 4 to the plasma membrane requires CD 147 ( basigin ) with which they remain tightly associated . ^^^ Here we demonstrate by site directed mutagenesis that removal of all accessible cysteine residues on MCT 4 does not prevent this inhibition . pCMBS treatment of cells abolished co immunoprecipitation of MCT 1 and MCT 4 with CD 147 and enhanced labeling of CD 147 with a biotinylated thiol reagent . ^^^ These data imply that inhibition of MCT 1 and MCT 4 activity by pCMBS is mediated through its binding to CD 147 , whereas MCT 2 , which associates with gp 70 , is insensitive to pCMBS . ^^^

In addition , expression of chaperone protein CD 147 , which is needed for the activity of MCT 1 and MCT 4 , was studied in the rumen of suckling calves . ^^^ Developing rumen of suckling calves expressed MCT 1 and MCT 4 and positive correlation was found between MCT 1 and CD 147 . ^^^
