Activation of type 1 metabotropic glutamate receptors ( mGluRs ) with DHPG ( 3 , 5 dihyidroxy phenylglycine ) activates PKC gamma but not PKC alpha or beta . 0.60107478^^^

Using subcellular fractionation and Western blotting with isoform specific antibodies and immunocytochemical localization with confocal laser scanning microscopy , we observed that phorbol ester and glutamate have different effects on PKC isoform redistribution : Whereas phorbol ester resulted in translocation of PKC alpha and PKC gamma toward a membrane fraction , the glutamate mediated rise in intracellular calcium concentration induced a translocation mainly toward a detergent insoluble , cytoskeletal fraction . ^^^ Immunocytochemical analysis revealed an isoform specific translocation following glutamate treatment : PKC gamma was translocated mainly to cytoplasmic , organelle like structures , whereas PKC alpha redistributed to the plasma membrane and into the cell nucleus . ^^^

The data suggest that PKCalpha / beta and PKCgamma are activated during different post tetanic phases and metabotropic glutamate receptor activation might be essential for tetanus induced translocation of postsynaptic PKCgamma only . . ^^^

Recent data in the literature suggest that it inhibits glutamate accumulation occurring during ischemia and the translocation of protein kinase C gamma ( PKC gamma ) . ^^^

RESULTS : Glutamate stimulation of astrocytes triggered highly localized GFP PKCgamma plasma membrane translocation events , induced rapid oscillations in GFP PKCgamma translocation , and generated GFP PKCgamma translocation waves that propagated across and between cells . ^^^ Interestingly , GFP conjugated C 1 domains from PKCgamma or PKCdelta that have been shown to bind diacylglycerol ( DAG ) also oscillated between the cytosol and the plasma membrane after glutamate stimulation , suggesting that PKC is repetitively activated by combined oscillating increases in Ca ( 2+ ) and DAG concentrations . ^^^

In this article , we investigate PKCgamma translocation in hippocampal neurons and show that electrical or glutamate stimulation leads to a striking enrichment of PKCgamma in synaptic spines and dendritic branches . ^^^

Here , we investigate the role of ionotropic and metabotropic glutamate receptor ( iGluR and mGluR , respectively ) activation on the generation of Ca2+ and diacylglycerol ( DAG ) signals and the subsequent activation of the neuron specific PKCgamma isoform in hippocampal neurons . ^^^

Ankle joint chondrocytes appeared to show significant differences in levels of the integrin associated proteins CD 98 , CD 147 and galectin 3 , PKCgamma and differences in responses to glutamate were seen . ^^^
