| Here , we report the isolation and characterization of a SNAP 25 interacting protein that is the rat homologue of mouse hepatocyte growth factor regulated tyrosine kinase substrate ( Hrs ) . ^^^ Hrs interacts with SNAP 25 and regulates Ca ( 2+ ) dependent exocytosis . ^^^ Hrs specifically interacts with SNAP 25 , but not SNAP 23 / syndet . ^^^ The association of Hrs and SNAP 25 is mediated via coiled coil interactions . ^^^ Using an Hrs specific antibody , we have shown that Hrs is highly enriched in brain , where it codistributes with SNAP 25 in most brain regions . ^^^ |

| A region of Hrs predicted to form a coiled coil required for binding the Q SNARE , SNAP 25 , mimicked the inhibition of endosome fusion produced by full length Hrs , and was sufficient for endosome binding . ^^^ SNAP 25 , syntaxin 13 , and VAMP 2 were bound from rat brain membranes to the Hrs coiled coil domain . ^^^ Hrs inhibited formation of the syntaxin 13 SNAP 25 VAMP 2 complex by displacing VAMP 2 from the complex . ^^^ These data suggest that SNAP 25 is a receptor for Hrs on early endosomal membranes and that the binding of Hrs to SNAP 25 on endosomal membranes inhibits formation of a SNARE complex required for homotypic endosome fusion . . ^^^ |

| INTERVENTIONS : Incubation of platelets with different concentrations ( 30 microM , 100 microM , 500 microM , 1000 microM ) of the nitric oxide donor S nitroso N acetyl d , l penicillamine ( SNAP ) for varying incubation times ( 0 hrs , 1 hr , 2 hrs , 4 hrs ) with and without endothelial cells . ^^^ Time course studies with 30 microM of SNAP showed an inhibitory effect only after 0 , 1 , and 2 hrs of incubation , whereas after 4 hrs of incubation the inhibition of platelet aggregation could not be detected any more . ^^^ Endothelial cells significantly increased the inhibitory effect of SNAP after 1 and 2 hrs of incubation . ^^^ |
