A novel human nm23 / nucleoside diphosphate ( NDP ) kinase gene , called nm 23 H4 , was identified by screening a human stomach cDNA library with a probe generated by amplification by reverse transcription polymerase chain reaction . ^^^ The full length cDNA sequence predicts a 187 amino acid protein possessing the region homologous to NDP kinases with all residues crucial for nucleotide binding and catalysis , strongly suggesting that Nm 23 H4 possesses NDP kinase activity . ^^^ Compared with these proteins , Nm 23 H4 contains an additional NH 2 terminal region that is rich in positively charged residues and could indicate routing to mitochondria . ^^^ The nm 23 H4 gene has been localised to human chromosomal band 16p13 . 3 . ^^^ Nm 23 H4 naturally possesses the Pro Ser substitution equivalent to the K pn mutation ( P97S ) of Drosophila . . ^^^

Overexpression of nm 23 H4 RNA in colorectal and renal tumours . ^^^ A new member of the nm 23 gene family , designated nm 23 H4 , was cloned recently . ^^^ Nm 23 H4 is a mitochondrial protein . ^^^ To determine whether nm 23 H4 could have a function in tumour progression like nm 23 H1 or nm 23 H2 , we analysed nm 23 H4 expression in 18 renal tumours and 42 colorectal carcinomas obtained from patients who underwent curative resection . ^^^ As compared to the corresponding healthy tissue , 14 renal tumours and 41 colorectal carcinomas showed increased nm 23 H4 mRNA expression . ^^^

Expression of the nm 23 homologues nm 23 H4 , nm 23 H6 , and nm 23 H7 in human gastric and colon cancer . ^^^ Whether nm 23 H4 , H 6 , and H 7 play a role in tumours is , however , largely unknown . ^^^ Increased expression of these genes , most strikingly nm 23 H4 and H 7 , was observed in the majority of tumours analysed . ^^^ In contrast , by immunohistochemical analysis , nm 23 H4 and H 6 overexpression correlated with the intestinal tumour type in gastric cancer tissues , whereas no increased immunoreactivity for the three nm 23 proteins was noted in the diffuse type tumour specimens . ^^^ These findings indicate that nm 23 H6 , and particularly nm 23 H4 and H 7 , may be involved in the development of colon and gastric carcinoma , the latter possibly in a type specific manner . ^^^
